Scheduling Equipment
To use the DNA Sequencing Core Lab, contact Hector Rivera at 626-256-HOPE (4673), ext. 63349 or hrivera@coh.org or Harry Gao at 626-256-HOPE (4673), ext. 65316 or hgao@coh.org.

If you are a first time user of our facility, we do ask you to set up a 20-minute training session with us prior to your first sample submission to set up a new account and familiarize you with our online service. DNA samples are submitted purified using commercially available kits and checked for impurities, contamination and concentration.

We encourage you to run your samples (uncut) on an agarose gel to confirm purity before submission.

For plasmid DNA, we request 200 ng of total DNA sample at a concentration of 100 ng/ul.
For BAC clones we request between 3-5 ug of DNA total at a concentration of 1 ug/ul.

Custom primers at a concentration of 1 picomole/ul are supplied by the user. General sequencing primers are available in the lab. Other information on the size of the insert, PCR product and vector is required.

Turn-around Time
City of Hope Cancer Center members have priority over non-members and outside users. Normally, 24-48 hours are required for analyses. We have a cut-off time of 2 p.m. (PST) for samples requiring 24 hour data results. Samples received after 2 p.m. (PST) will be sequenced and processed the next business day.

Investigators will receive an email notice acknowledging their data is ready to be retrieved from our server.

Investigators can print sequence data and chromatograms from their own computers. We do not print sequence data unless it is a unique and non-recurrent situation.

All data are archived on DVDs and stored for users in the DNA Sequencing Lab unless requested otherwise. The data on the server is ghosted by Information Technology Services and backed by the Department of Information Sciences.

Quality Control
Standard Applied Biosystems maintenance procedures are carried out each day.

pGEM plasmid controls are routinely sequenced and analyzed for quality control. Generally 900 - 950+ bases of high quality sequence data are achieved.
