CyTOF-Helios (Fluidigm) utilizes mass cytometry, which uniquely combines time-of-flight mass spectrometry with metal-tagged antibodies or reagents to detect up to 50 simultaneous parameters on a per cell basis. Cellular targets (applicable to both intra- and extracellular) are labeled with metal-tagged antibodies, which are detected and quantified by time-of-flight mass spectrometry. Mass cytometry empowers researchers to interrogate simultaneously cellular phenotypes, function and signaling status in higher dimensions at the single-cell level. The high purity and choice of metal isotopes provide minimal background noise from signal overlap or endogenous cellular components. Current instrumentation has 135 available detection channels to ensure an ongoing ability to add more parameters.
This technology is applicable to provide deep profiling of translational and clinical research samples across a range of cell surface and intracellular markers.
