X-Ray Crystallography
The X-Ray Crystallography Core facility provides protein expression, biophysical characterization and structural determination to the scientists at City of Hope.

Protein expression services include initial analysis, generation of expression constructs, and expression in E. coli, yeast, insect and mammalian cell lines. Purification services include FPLCs with preparative grade affinity, ion-exchange, and size exclusion columns.
Instruments for biophysical characterization include:
- Biacore T200
- Water nanoITC
- Beckman-Coulter XLI Analytical Centrifuge
- Sapidyne Kinexa
- Jasco810 Circular Diochroism spectrophotometer
Instruments for structure determination include:
- Mosquito crystallization Robot,
- Two Forumatrix imaging robots
- Rigaku 007HF with an R-avis-IV++ imaging plate
- Oxford cryostream
In addition, the core has ample access to synchrotron sources, including SSRL and NSLS-II. Finally, the core subscribes to SBGrid and thus has all the software needed for structure determination and analysis.

- Mosquito Crystallization robot – Used to set up crystallization trials
- Formulatrix imagers – Used to image crystallization trials automatically
- Waters NanoITC – Used to measure thermodynamic properties (△H, △S and KD)
- Biacore T200 – Used to measure kinetics (protein-protein, small molecule-protein, DNA or RNA, etc.)
- Kinexa3000 – Used to measure ultra-high affinity interactions
- Jasco 815 CD or fluorescence polarization anisotropy – used to measure the secondary structure and hydrodynamic values
- Beckman Proteome Lab – Used to measure hydrodynamic interactions, including oligomerization and self-association constants
In a given year, City of Hope conducts more than 400 clinical trials enrolling more than 6,000 patients.
